• We generated 2nd–3rd generation explant human skin equivalents (Ex-HSEs) from 4mm skin biopsies. • 2nd–3rd generation Ex-HSEs have similar morphology and differentiation to 1st generation Ex-HSEs and native human skin. • Stratum corneum of 2nd and 3rd generation Ex-HSEs show the main SC lipid classes and long periodicity phase as in native human skin. • 2nd–3rd generation Ex-HSEs have more unsaturated free fatty acids and esterified ω-hydroxy ceramides compared to native human skin. |
