In oblique sections, ovals with a diameter of 800-900 A occur. The mitochondria are surrounded by a 100 A thick double outer membrane. The arrangement of the branching inner membranes is complicated. The cells contain organelles with a fine structure resembling that of crystallized substances. The cytoplasm contains typical α-cytomembranes.The spores of D. purpureum, D. sphaerocephalum and Acytostelium show the same double walls as earlier found in D. discoideum [25].The sorophore sheath in Dictyostelium is double [13] and the inner layer is probably formed inside the stalk in connection with cell wall formation. The stalk in Acytostelium consists of parallel fibril bundles. No real lumen exists within the stalk. X-ray analyses of the untreated stalk substance do not show cellulose reflections. After purification with hot aqueous 1 per cent sodium hydroxide, the stalk substance gives a X-ray diffraction pattern with reflections characteristic for native cellulose. The width of the cellulose fibrils is about 100 A. From a comparison between Acytostelium and Dictyostelium it is concluded that the stalk of Acytostelium and the sorophore outer sheath layer of Dictyostelium most probably are homologous. | ||

The ultrastructure of cells and cellulose membranes in Acrasiae |
abstract |
The ultrastructure of the cells and cellulose membranes in Acrasiae has been studied by means of electron microscopy using ultrathin sections of Dictyostelium discoideum, D. purpureum, D. sphaerocephalum and Acytostelium leptosomum. The vacuoles of the myxamoebae contain multi-layered configurations, possibly waste products formed from the bacteria. The cell membrane and the vacuole membrane are double lined with a total thickness of 80-90 A.The nuclear membrane is double. |
