Under these conditions a complete breakdown of the ΔpH was observed in the presence of 50 mM KCl. The results are interpreted in such a way that in isolated vacuoles the maintenance of the ΔpH is mainly based on the low permeability of the tonoplast membrane for protons, while in the presence of ionophores a donnan equilibrium is established. The main advantages of the 9-AA method are: (i) separation of the vacuoles from the suspension medium is not required and (ii) it allows rapid and continuous monitoring of changes of the ΔpH. | ||

It is suggested that binding of 9-AA molecules to membrane surfaces and/or proteins contributes only little to the fluorescence quenching. Addition of carbonylcyanide-p-trifluoromethoxy-phenyl-hydrazone (FCCP) caused little decline of the ΔpH. In the absence of ionophores the ΔpH was relatively insensitive to the addition of external salt but became highly sensitive in the presence of either valinomycin or valinomycin plus FCCP. Under these conditions a complete breakdown of the ΔpH was observed in the presence of 50 mM KCl. The results are interpreted in such a way that in isolated vacuoles the maintenance of the ΔpH is mainly based on the low permeability of the tonoplast membrane for protons, while in the presence of ionophores a donnan equilibrium is established. The main advantages of the 9-AA method are: (i) separation of the vacuoles from the suspension medium is not required and (ii) it allows rapid and continuous monitoring of changes of the ΔpH. |

The fluorescent amine 9-aminoacridine (9-AA) has been used to investigate the proton concentration difference (ΔpH) across the tonoplast membrane of mesophyll vacuoles isolated from Valerianella locusta (L.). Fluorescence quenching was measured in dependence on the concentration of isolated vacuoles, the pH of the suspending medium and the concentration of the amine added. An intravacuolar pH of about 5.0 was calculated at an external pH of 6.9-7.6.
